Rapid and high-throughput identification of four kinds of G+ bacteria
G+ bacteria are now the leading cause of hospital-acquired infections, and their resistance to antibiotics has emerged in the last decade as a major clinical problem. CapitalBio G+ Bacteria Identification and Drug Resistance Detection Array Kit was developed for the rapid and high-throughput identification of four kinds of G+ bacteria, including Staphylococcus aureus, coagulase-negative staphylococcus, Enterococcus faecalis and Enterococcus faecium, which cover up to 90 percent of the most prevalent G+ pathogenic isolates found in clinical specimens. Simultaneously, the kit can be used to determine the following five antibiotic resistance genes in the test clinical isolate:
mecA found in MRS (Methicillin-Resistant Staphylococcus)
ermA gene and ermC found in MLS (Macrolide-Lincosamide-Streptogramin B) Resistant Staphylococcus B
msrA found in Macrolide-Resistant Staphylococcus
aac(6´)-aph(2´´) found in both HLAR (High-Level Aminoglycoside Resistant) Enterococcus and Aminoglycosideresistant Staphylococcus
Principle
After a simple and rapid DNA extraction procedure, target bacterial gene fragments are specifically amplified if present in the isolate via a multiplex asymmetric PCR and are labeled with a fluorescent dye. The labeled DNA amplicons are then hybridized to a set of specific probes on the DNA chip. According to the positions of the positive probe signals, both the presence of target genes and their sequences can be inferred. Thus, both the particular bacterial infection can be identified and drug resistance can be detected.
