CapitalBio Antinuclear Antibody Profile Test Chip Kit is a protein array-based ANA detection kit that simultaneously tests for 8 different IgG antinuclear antibodies against SSA 52, SSA 60, SSB, Sm, RNP 68, Scl-70, Jo-1 and centromere B in the patient´s serum. The test has high sensitivity and specificity for ANA profiling and is a valuable aid for the diagnosis and prognosis of autoimmune diseases. Each chip has 12 separate test sub-arrays.
Systemic autoimmune diseases, such as systemic lupus erythematosis (SLE), rheumatoid arthritis, mixed connective tissue disease (MCTD), Sjogren´s syndrome (SS), systemic sclerosis (SSc), dermatomyositis 4(DM), polymyositis (PM) and CREST syndrome are a group of diseases characterized by the presence of antinuclear antibodies (ANA) circulating in a patient´s blood which attack the individual´s own body tissues. This group of diseases collectively affects 2-3% of the worldwide population.

ANA tests are routinely conducted in clinical laboratories. The results of the tests in conjunction with clinical symptoms and other laboratory findings assist physicians in the diagnosis and staging of systemic autoimmune diseases. The diseases have complex aetiologies, with one autoantibody found positive in several different diseases, while another disease may have several positive autoantibodies. The occurrence and quantities of autoantibodies may also vary in the same individual at different disease stages.

Assay Principles
Chips with immobilized antigens (SSA 52, SSA 60, SSB, Sm, RNP 68, Scl-70, Jo-1, centromere B) react with antinuclear antibodies in the patient´s serum. Unbound autoantibodies are removed by subsequent washing steps. An anti-human IgG conjugated with fluorescence dye is then applied to each block on the chip and any unbound conjugates are washed off. The fluorescence signals remaining on the chip are quantified by a microarray scanner (CapitalBio LuxScan™ 10K). The data obtained from the scanner are then analyzed by the ANA test system software to generate the test reports.
